355 Sensory neuronal STAT3 is critical for IL-31 receptor expression and inflammatory itch

چکیده
IL-31 receptor blockade suppresses pruritus of atopic dermatitis. However, cell type-specific contributions to itch, its expression mechanism, and the downstream signaling pathway induce itch remain unknown. Here, using conditional knockout mice, we demonstrated that IL-31-induced required sensory neuronal STAT3. was decreased in STAT3 deficient indicating dependent on neurons. In addition, an acute treatment with inhibitor wild type mice ameliorated without reducing expression, which suggested activation important for itch-inducing receptor. A cutaneous injection induced nuclear accumulation activated first neurons abundantly expressed then other itch-transmitting Consistently, found enhanced not only histamine-induced transmitted by receptor-expressing but also chloroquine-induced subsets. Finally, associated dermatitis partially strongly Thus, is essential further contributes IL-31-independent inflammatory itch.

A sensory neuron-expressed IL-31 receptor mediates T helper cell-dependent itch: Involvement of TRPV1 and TRPA1.
BACKGROUND Although the cytokine IL-31 has been implicated in inflammatory and lymphoma-associated itch, the cellular basis for its pruritic action is yet unclear. OBJECTIVE We sought to determine whether immune cell-derived IL-31 directly stimulates sensory neurons and to identify the molecular basis of IL-31-induced itch. METHODS We used immunohistochemistry and quantitative real-time PCR...
متن کاملIL-31 expression by inflammatory cells is preferentially elevated in atopic dermatitis.
Interleukin-31 (IL-31) is a recently discovered cytokine expressed in many human tissues, and predominantly by activated CD4(+) T cells. IL-31 signals through a heterodimeric receptor consisting of IL-31 receptor alpha (IL-31RA) and oncostatin M receptor beta (OSMR). Earlier studies have shown involvement of IL-31 and its receptor components IL-31RA and OSMR in atopic dermatitis, pruritus and T...

متن کاملCritical role for STAT3 in IL-17A-mediated CCL11 expression in human airway smooth muscle cells.
IL-17A has been shown to be expressed at higher levels in respiratory secretions from asthmatics and to correlate with airway hyperresponsiveness. Although these studies raise the possibility that IL-17A may influence allergic disease, the mechanism remains unknown. We previously demonstrated that IL-17A mediates CC chemokine (CCL11) production from human airway smooth muscle (ASM) cells. In th...
